If it is not on the prescription bottle on how to take it, get ahold of your doctor and ask?

I have the same prescription and can tell you what doctor and pharmacist told me:
Estrace is one in the morning and the 2nd one at night.
Spiro is once in the morning.
I was told to take them on a full stomach/with food.
Was going to try the morning one by just swallowing it and the one at night under the tongue.

I'm on Spironolactone myself.  My physician told me that I could take it at any time of the day, but kind of gave me a hint so far as the consequences of each time.  It has two very prominent negative effects:

So if you take it in the morning, you'll more likely notice that you are very tired.  If you take it later at night, then you might notice that you need to make a lot of trips to the restroom (it'll disrupt or discomfort you as you try to sleep).  Of course: Always consult with your physician.  I am not a medical expert.

Personally, I like to take it at night, and taking it around 6 pm makes it most manageable for myself.

Ya she told me to stay hydrated she says Spiro is a anti something i cant name off the top of my head. Just wow can't believe I'm finally starting. i will probably take mine mid day since i work mornings. but i should probably take the estrace then to for best effect i would think. guhhh I can't wait to see her again next month so i can actually ask her all the question i forgot to. I'm starting a list of them lol.

Antimineralocorticoid (anti-aldosterone), thus increasing sodium excretion and potassium retention. You also need to eat more salt and limit high potassium stuff. Moderate intake of diuretics.

Both Estrace and Spiro have a short half-life, around 13-17 hours, if I recall, so usually recommended to take twice daily. Best to check with your doctor.
