Let me see: wallet, checkbook, make up bag, perfume, mirror, box of q-tips, 2 combs, small mirror, band aid box, tylenol bottle, assorted pens and pencils, caffeine pills, flash light, Swiss army knife, compass, whistle, 2 tweezers, forceps, can of candy, tooth brush, floss, 2 tampons, and my trusty colt

Well, with that can of candy, you might also want to consider carrying toothpaste.  :P

By the way, VM, hats, cute or otherwise, go on your head, not your purse.

Anyway, I never liked purses. I prefer backpacks since they're easy to ride a bike with and they keep the weight on both shoulders instead of one. Here's what's in mine.

-Wallet.
-Phone.
-Toothbrush to clean gunk out of my bike chain.
-Bike repair kit.
-Pencils.
-A pen.
-Snacks.
-Bandages.
-Flashlight/glowstick/whistle combo. (I know, it's awesome  ;D)

Ladies, you might want to consider placing your pepper spray in a more accessible place that can be reached quickly. It would be really bad if you reached into the bottomless abyss that is a woman's purse and pulled out your can of candy instead of your can of pepper spray. Yikes.  :(

My first line of defense is me. The people I don't want to send to the hospital I use the pepper spray on. They are the lucky ones.

I don't eat candy and my purse, backpack or whatever I'm using at the time tends to be well organized so I know exactly where all of the contents are located

Oh, that toothpaste comment was directed at peky. It was a joke.

A place for everything and everything in its place.

I do keep a candy bar and a glucose tab in my purse, but they are "in their place".

I can find everything in the dark.

I keep candy in mine for my son. It keeps him quiet when I need him to stop talking.  ;D

From what I've learned, though, carrying things like a pepper spray isn't the best of ideas, because they can easily be used against you and make matters worse.

My purse side grew over time. I had a small green purse with only my wallet and keys in it. But further I got into HRT my skin started drying and getting itchy. Moisturizers were needed. I just slowly filled it with things I need. When it over flowed got a bigger one. It is moving back the other way as I don't need a bigger one. Wallet, keys, MP3 player, 2 pads to write on, Makeup which will change for me in the summer won't need it or as much I should say. I do seem to have on hand things like pain relievers were as before I was ask supervisor for them. A lot of change that I through in it which is great to have when I need it. pins to write with, jewelry that I may have taken of. Ponytail holders a round brush. Before I used a purse I would wear a fanny pack I did not like putting things in my pockets. I have grown attached to it and it pretty much goes every were I do. If I for get it I feel naked with out it.
